چکیده
Background With pathway-specific trials in PD associated with variants the glucocerebrosidase gene (PDGBA) under way, we need markers that confirm impact of genetic patient-derived biofluids order to allow patient stratification merely based on genetics and might serve as biochemical read-out for target engagement. Objective To explore GBA-pathway-specific biomarker profiles cross-sectionally (TUEPAC-MIGAP, PPMI) longitudinally (PPMI). Methods We measured enzyme activity lysosomal glucocerebrosidase, CSF levels glucosylceramides (upstream substrate glucocerebrosidase), ceramides (downstream product lactosylceramides, sphingosines, sphingomyelin (by-products) total ?-synuclein PDGBA patients compared PDGBA_wildtype patients. Results Cross-sectionally both cohorts PPMI: (1) was significantly lower PDGBA_wildtype. (2) upstream substrates (glucosylceramides species) were higher (3) All these findings most pronounced severe mutations (PDGBA_severe). TUEPAC-MIGAP PPMI, downstream-products (ceramides) PDGBA_severe. by-products sphinganine sphingosine-1-phosphate PPMI species lactosylceramides Interpretation These GBA have a relevant functional Bridging gap between now allows clinical mutation status. Importantly, all prominent variants. © 2021 International Parkinson Movement Disorder Society

All - Termination ( SCP )
We recently introduced the All-Termination(T ) problem: given a termination solver T and a function F , find every subset of the formal parameters to F whose consideration is sufficient to show, using T , that F terminates. These subsets can be harnessed by a theorem prover to locate and justify induction schemes, and are also useful for guiding rewriting heuristics and ensuring their terminati...
متن کاملImpact of SCP-2/SCP-x gene ablation and dietary cholesterol on hepatic lipid accumulation.
While a high-cholesterol diet induces hepatic steatosis, the role of intracellular sterol carrier protein-2/sterol carrier protein-x (SCP-2/SCP-x) proteins is unknown. We hypothesized that ablating SCP-2/SCP-x [double knockout (DKO)] would impact hepatic lipids (cholesterol and cholesteryl ester), especially in high-cholesterol-fed mice. DKO did not alter food consumption, and body weight (BW) ...
متن کاملSCP-2/SCP-x gene ablation alters lipid raft domains in primary cultured mouse hepatocytes.
Although reverse cholesterol transport from peripheral cell types is mediated through plasma membrane microdomains termed lipid rafts, almost nothing is known regarding the existence, protein/lipid composition, or structure of these putative domains in liver hepatocytes, cells responsible for the net removal of cholesterol from the body. Lipid rafts purified from hepatocyte plasma membranes by ...
متن کاملThree-dimensional structure/function analysis of SCP-2-like2 reveals differences among SCP-2 family members.
Mosquito sterol carrier protein-2 (AeSCP-2) and sterol carrier protein-2-like2 (AeSCP-2L2) are members of the SCP-2 protein family with similar expression profiles in the mosquito life cycle. In an effort to understand how lipids can be transported by different SCP-2 proteins, the three-dimensional crystal structure of AeSCP-2L2 was solved at 1.7 A resolution. AeSCP-2L2 forms a dimer and binds ...
متن کاملSCP in Agriculture and Rural Development
Introduction Achieving more sustainable consumption and production in the agricultural sector requires a coherent response to the intimate linkages between agricultural production, agro-ecosystems and the people who rely on them, and also the concerns and preferences of a growing number of consumers. Less input intensive and more resource efficient agriculture offers a means to strengthen the c...
